SS-31 (Elamipretide) is a mitochondria-targeting tetrapeptide designed to selectively bind cardiolipin in the inner mitochondrial membrane. It is widely used in research focused on mitochondrial function, oxidative stress, and cellular energy metabolism. SS-31 enhances mitochondrial bioenergetics, reduces reactive oxygen species (ROS) production, and stabilizes mitochondrial structure, making it a valuable tool in studies of aging, metabolic disorders, and tissue injury.

Questions
SS-31 Common Questions
What’s pIC50?
pIC50 is the negative log of the IC50 value when converted to molar. That is, pIC50=-log(IC50).
What’s Kd?
Dissociation consent (Kd) reflects the affinity of a compound for its target. In some cases, it can be equivalent to Ki.

Targets & IC50
Targets:
Cardiolipin
Mitochondrial Respiration
Reactive Oxygen Species Regulation
IC50:
Not applicable (SS-31 acts via membrane binding and mitochondrial modulation, not classical inhibition)
In vitro
METHODS: Cardiomyocytes, neuronal cells, and endothelial cells were treated with SS-31 (0–10 μM) for 24–72 hours. Mitochondrial function, ROS production, and ATP levels were assessed using fluorescence assays, oxygen consumption rate (OCR) analysis, and Western blot detection of mitochondrial proteins.
RESULTS: SS-31 improves mitochondrial respiration and increases ATP production while reducing reactive oxygen species (ROS) levels. It stabilizes mitochondrial membranes by binding cardiolipin and enhances cellular resistance to oxidative stress. SS-31 demonstrates protective effects in multiple cell types, supporting improved bioenergetic efficiency and cell survival. No significant cytotoxicity is observed at commonly used experimental concentrations. [1]
