In vitro
METHODS: Rat pituitary cell lines and primary anterior pituitary cells were treated with CJC-1295 Without DAC (0–100 nM) for 1–24 hours. Growth hormone secretion and downstream signaling were evaluated using ELISA assays and cAMP response measurements.
RESULTS: CJC-1295 Without DAC stimulates growth hormone secretion in a dose-dependent manner via activation of the GHRH receptor. It enhances intracellular cAMP levels and promotes pulsatile GH release. Compared to native GHRH, the modified peptide demonstrates improved stability while maintaining receptor specificity. No significant cytotoxicity is observed at commonly used experimental concentrations. [1]
