LL-37 is a human antimicrobial peptide belonging to the cathelicidin family and is the only cathelicidin-derived peptide identified in humans. It plays a critical role in innate immunity, exhibiting broad-spectrum antimicrobial activity against bacteria, viruses, and fungi. In addition to its antimicrobial properties, LL-37 is involved in immune modulation, wound healing, and regulation of inflammatory responses, making it widely used in research related to host defense and tissue repair.

Questions
LL37 Common Questions
What’s pIC50?
pIC50 is the negative log of the IC50 value when converted to molar. That is, pIC50=-log(IC50).
What’s Kd?
Dissociation consent (Kd) reflects the affinity of a compound for its target. In some cases, it can be equivalent to Ki.

Targets & IC50
Targets:
Bacterial Cell Membrane
Formyl Peptide Receptor 2 (FPR2)
Innate Immune Response
NF-κB Signaling Pathway
IC50:
Not applicable (LL-37 acts via membrane disruption and receptor-mediated signaling, not classical inhibition)
In vitro
METHODS: Human keratinocytes, macrophages, and bacterial cultures (e.g., E. coli, S. aureus) were treated with LL-37 (0–50 μM) for 6–48 hours. Antimicrobial activity, cytokine expression, and cell migration were evaluated using MIC assays, ELISA, and wound healing assays.
RESULTS: LL-37 exhibits broad-spectrum antimicrobial activity by disrupting microbial membranes. It modulates immune responses through interaction with receptors such as FPR2 and regulates inflammatory signaling pathways including NF-κB. Additionally, LL-37 promotes wound healing by enhancing keratinocyte migration and angiogenesis. No significant cytotoxicity is observed at controlled experimental concentrations. [1]
