KPV (Lys-Pro-Val) is a naturally derived tripeptide fragment of α-melanocyte-stimulating hormone (α-MSH). It is widely studied for its anti-inflammatory and immunomodulatory properties, particularly in pathways associated with cytokine regulation and innate immune responses. KPV has been used in research involving skin inflammation, gastrointestinal disorders, and tissue repair due to its ability to modulate inflammatory signaling without significant systemic effects.

Questions
KPV Common Questions
What’s pIC50?
pIC50 is the negative log of the IC50 value when converted to molar. That is, pIC50=-log(IC50).
What’s Kd?
Dissociation consent (Kd) reflects the affinity of a compound for its target. In some cases, it can be equivalent to Ki.

Targets & IC50
Targets:
NF-κB Signaling Pathway
Tumor Necrosis Factor-alpha (TNF-α)
Interleukin-6 (IL-6)
IC50:
Not applicable (KPV is a regulatory peptide, not a classical enzyme inhibitor)
In vitro
METHODS: Human keratinocytes, macrophages, and intestinal epithelial cell lines were treated with KPV (0–100 μM) for 24–72 hours. Inflammatory cytokine expression and signaling pathway activity were evaluated using ELISA, qPCR, and Western blot analysis.
RESULTS: KPV significantly reduces the expression of pro-inflammatory cytokines such as TNF-α and IL-6 by modulating NF-κB signaling pathways. It demonstrates anti-inflammatory effects in both skin and epithelial cell models and supports cellular recovery under inflammatory stress conditions. No significant cytotoxicity is observed at commonly used experimental concentrations. [1]
